Expression of hypoxia inducible factor-1 alpha in pediatric nephroblastoma
- Abstract:
- Objetive The aim of this study was to explore the expression of hypoxia-inducible factor-1α(HIF-1α)in children with wilms tumor(WT)and its correlation with clinicopathology, microvessel density(MVD)and prognosis. Methods 45 cases with nephroblastoma resecting,which specimens were confirmed after surgical pathologically in our unit from January 2008 to January 2012,and they were divided into five groups by age, sex, histopathological type, clinical stage and NWTS-5 risk factors, respectively. The expression of HIF-1α and CD 34 in WT organizations and adjacent tissues was tested by immunohistochemical method and MVD was calculated by CD 34 staining,which were analysis combined with the clinical data and results of the follow-up. Results The ratio of HIF-1α expressed in WT group was higher than adjacent tissues group(95.6% vs 40%,P<0.05)and intensity was increased with the increasing of clinic stages of the tumor(P<0.05). MVD was higher in WT group than in adjacent tissue groups(58.22±10.009 vs 49.43±8.114,P<0.05)and had a positive correlation to HIF-1α(P<0.05). HIF-1α expressed in high risk group was higher than in low risk group(P<0.05)and the postoperative cumulated survival rate was higher in low expression of HIF-1α group than in the high expression group(P<0.05). Conclusions High expression of HIF-1α is correlated to the development of WT and improves angiogenesis, which indicates a poor prognosis.
